Surprisingly, we found that Thr-Gly-S-adenosylmethionine (SAM) metabolism consistently showed the best correlation with pluripotency. 13Carbon isotope metabolomics further revealed that Thr was catabolized to generate Gly and acetyl-CoA, and ultimately SAM - essential for all methylation reactions. Thr is required for SAM and histone H3K4 methylation in mouse ESCs, thus regulating the open euchromatin and pluripotency of ESCs. Our study shed light on a novel amino acid pathway in stem cells, and demonstrated that metabolic conditions can direct cell fate. In summary, my work has helped us to understand how we can reprogram and manipulate metabolic networks to regulate stem cell homeostasis.

For an increasing number of cancers, the cell of origin has been demonstrated to be the resident adult stem cell. One such cancer is squamous cell carcinoma, for which recent studies in our lab traced its origin to the hair follicle stem cells. Malignant transformation is thought to coincide with a dramatic shift towards the use of glycolysis and establishment of a 'Warburg' state - increased metabolism of glucose to lactate. How the Warburg Effect is established during tumor initiation and progression in vivo remains unclear. The current consensus is that the bulk of the energy generated in most adult tissue cells is created by oxidative phosphorylation, while more highly proliferative cells, such as activated immune cells and cells transformed to make a tumor, mainly use glycolysis. Little is known about how individual cell types generate energy in vivo, however, and how their metabolism influences basic cell fate decisions such as cell division, migration or differentiation. Using genetically engineered mouse models that allow the study of both tissue homeostasis and the Warburg Effect in vivo, I have made important observations that provide the basis for new investigations into the role of metabolism in key cell fate decisions by adult stem cells. In this dissertation I present data indicating that hair follicle stem cells possess a unique metabolic profile that may be critical for their maintenance and for their response to oncogenic insults. Importantly, they suggest the possibility that the "Warburg Effect" is the result of the expansion of an already glycolytic subpopulation, namely the hair follicle stem cells.

The rate of glycolytic metabolism changes during differentiation of human embryonic stem cells (hESCs) and reprogramming of somatic cells to pluripotent stem cells. However, the functional contribution of glycolytic metabolism to pluripotency is unclear. Here we show that the degree of pluripotency is associated with glycolytic rate, whereby naive hESCs exhibit increased glycolytic flux, MYC transcriptional activity, and nuclear localization of N-MYC relative to primed hESCs. This is consistent with the inner cell mass of human blastocysts which exhibit increased MYC transcriptional activity relative primed hESCs and elevated nuclear N-MYC levels. Reduction of glycolysis decreases self-renewal of naive hESCs and feeder-free cultured primed hESCs, but not primed hESCs grown in feeder-supported conditions. Reduction of glycolysis in feeder-free primed hESCs also enhances neural specification. These findings reveal associations between glycolytic metabolism and the state of pluripotency, differences in the metabolism of feeder- versus feeder-free cultured hESCs, and identify methods for regulating self-renewal and initial cell fate specification of hESCs.

This volume contains the Proceedings of the Serono Symposium on Pre implantation Embryo Development, held in Newton, Massachusetts, in 1991. The idea for the symposium grew out of the 1989 Serono Symposium on Fertilization in Mammals* at which preimplantation development was the predominant suggestion for a follow-up topic. This was indeed a timely subject in view of the recent resurgence of interest in this funda mental phase of embryogenesis and its relevance to basic research and applied fertility studies in humans, food-producing animals, and endangered species. The symposium brought together speakers from a broad range of disciplines in order to focus on key regulatory mechanisms in embryo development, using a wide variety of animal models, and on representative topics in human preimplantation embryogenesis. The culmination of preimplantation development is a blastocyst con taining the first differentiated embryonic tissues and capable of initiating and sustaining pregnancy. The central objective of the symposium was to throw light on the regulation of cellular and molecular events underlying blastocyst formation. It was particularly appropriate that the date of the symposium marked the 20th anniversary of the publication of the classic volume Biology of the Blastocyst, the proceedings of an international workshop held in 1970. This book, which summarized most of the information then available on this topic in mammals, was edited by the pioneer in blastocyst research, Dr. Richard B1andau, who was the guest speaker at the symposium.
