This text assesses the role of dendritic cells, T and B lymphocytes, epithelial and mast cells, eosinophils, neutrophils, macrophages, and their products in the initiation and progression of airway inflammation, regulation, and repair. It unveils advances in immunology and cell biology to generate new therapeutic regimens for inflammatory and aller

This book explores the cellular and molecular mechanisms involved in asthma and allergic diseases, from basic immunology to the treatment of patients. Arising from a special symposium held to celebrate Professor A. Barry Kay's 20th anniversary as Head of the Department of Allergy and Clinical Immunology at the National Heart and Lung Institute, London, the publication presents 18 review articles by an impressive cross section of experts in the field. Topics considered include Th1 and Th2 cells, the regultion of Th2 cytokine transcription, the role of T cells in the immunopathogenesis of asthma, and MHC-restricted IgE-independent T-cell-induced late-phase responses in human subjects. Further chapters focus on eosinophil adhesion and activation, the role of C-C chemokines in the recruitment of eosinophils, and the interactions between eosinophils and fibroblasts. T-cell recruitment and T-cell interactions in cutaneous basophil hypersensitivity and fibrotic lung disease are discussed, as is the molecular pathology of allergic disease. Finally, treatment approaches are considered in chapters on the molecular basis of corticosteroid actions in asthma, immunosuppression in asthma, and immunotherapy of allergic diseases.

Allergic diseases are complex and involve a range of environmental factors interacting with a susceptible genotype. The familial clustering of diseases, such as asthma and hay fever, has been recognised for over two centuries, but identification of the genetic basis to this had to await the molecular biological revolution. Estimates of the contribution that genetic factors make to asthma susceptibility range from 35% to 70%. For the majority of allergic diseases, segregation analysis has not identified a consistent Mendelian pattern of inheritance, which, when combined with multiple phenotypes and environmental interactions, has made identifying candidate genes especially difficult and, at times, controversial. Part of the difficulty has been lack of agreement over phenotype definitions, reduced power of studies to predict linkage and association, and, importantly, lack of true heterogeneity between populations. Despite these difficulties, the last decade has witnessed enormous progress in this field.

Asthma is a chronic complicated airway disease whose mortality and morbidity rates have increased worldwide. Some common treatments for asthma cannot control it nor even prevent simple inflammation and relieve shortness of breath, so understanding its molecular pathways and pathophysiology is required in order to better understand the disease and to diagnose, control and treat it. Allergic Asthma Immunopathogenesis: Immunopathology of the Allergic Asthma presents a new glance to the allergic asthma pathophysiology by focusing on the immunopathogenesis and allergo-inflammation mechanisms of asthma and cell signaling pathways that can be used for various research purposes, including the design of anti-asthma treatment and target therapy for scientists and researchers. Covers new molecular pathophysiology mechanisms and cell signaling pathways in pathogenesis of allergic asthma that have been introduced in recent research Discusses immunologic basis of the allergy and hyperresponsiveness Includes results of recent research focusing on the diagnosis and treatment of allergic asthma

Eosinophils represent approximately 1% of peripheral blood leukocytes and play a central proinflammatory and immunoregulatory role in various immune disorders. This volume provides the reader with a comprehensive survey of the major aspects of human eosinophilic biology and immunology as well as the most clinically relevant aspects of eosinophil-related disorders. Expert contributions cover the ultrastructural and phenotypic characteristics of human eosinophils, and their fundamental biochemical features including receptors for IgE and chemokines. The production of cytokines and their role in the activation of eosinophils are reviewed. Furthermore, the classic eosinophil-related disorders such as hypereosinophilia, drug reactions, allergic disorders and some cardiovascular diseases are described. Each of the chapters is written by an investigator actively engaged in research on the topic under consideration. The well-edited and stimulating material compiled in this volume will be of interest to all those working in allergology, immunology, biochemistry and pulmonary medicine.

Severe asthma is a form of asthma that responds poorly to currently available medication, and its patients represent those with greatest unmet needs. In the last 10 years, substantial progress has been made in terms of understanding some of the mechanisms that drive severe asthma; there have also been concomitant advances in the recognition of specific molecular phenotypes. This ERS Monograph covers all aspects of severe asthma – epidemiology, diagnosis, mechanisms, treatment and management – but has a particular focus on recent understanding of mechanistic heterogeneity based on an analytic approach using various ‘omics platforms applied to clinically well-defined asthma cohorts. How these advances have led to improved management targets is also emphasised. This book brings together the clinical and scientific expertise of those from around the world who are collaborating to solve the problem of severe asthma.
