The effect of temperature on spinach seed germination was evaluated using a total of nine spinach genotypes and seven temperatures: 10, 15, 20, 25, 30, 32, and 35 oC in growth chambers. Genetic variation was observed. 'Donkey', 'Marabu', and 'Raccoon' showed higher seed germination percentage with over 70% at 30 and 32 oC, indicating the three spinach genotypes had heat-tolerance for germination. However, all spinach genotypes except 'Ozarka II' had lower germination percentages of less than 30% while 'Ozarka II' had 63% at 35 oC, indicating 'Ozarka II' may be a source of heat-tolerance for seed germination. Seed germination may be useful as an early screening method for heat-tolerant spinach genotypes. The whole-plant experiment was conducted using two temperatures, 20 and 32 oC, in growth chambers. Variation was observed among spinach cultivars for leaf area and shoot dry weight at 32 oC. 'Samish' was the only cultivar to have no significant difference between 20 and 32 oC for leaf area, while the leaf area for 'Ozarka II', 'Donkey', and 'Marabu' decreased from 20 to 32 oC. This may indicate that 'Samish' is heat-tolerant. However, significant differences were not observed among cultivars for shoot dry weight. The results from whole-plant growth at high temperature did not parallel those of the seed germination study, and the evidence does not indicate that seed germination can be used as a screening tool for heat tolerant cultivars. Bolting, tallness, and erectness are important morphological traits in spinach breeding. A total of 288 USDA spinach accessions were used as the association panel in this research. Single nucleotide polymorphisms (SNPs) discovered through genotyping by sequencing (GBS) were used for genotyping. Three SNP markers, AYZV01001038_398, AYZV01031624_1060, and AYZV01088923_95 were found to be associated with bolting. Eight SNP markers, AYZV01011130_540, AYZV01180397_2162, AYZV01069590_19842, AYZV01105690_376, AYZV01058838_64, AYZV01152613_2532, AYZV01113619_2197, and AYZV01003134_248 were associated with tallness. Four SNP markers, AYZV01137843_229, AYZV01158294_79, AYZV01023368_256, and AYZV01097131_197 were associated with erectness. These SNP markers may be used in spinach molecular breeding to select spinach bolting, tallness and erectness through marker-assisted selection.

When trying to solicit authors for this book it became apparent that the causal factors for heterosis at the physiological and biochemical level are today almost as obscure as they were 30 years ago. Though biometrical-genetical analyses point to dispersion of complementary genes - not overdominance - as the major cause of the phenomenon, plant breeders' experience still suggests a cautious, pragmatic approach to the dominance-overdominance controversy in breeding hybrid cultivars. Thus we are faced with a striking discordance between our limited comprehension of the causal factors and mechanism of heter osis on the one hand, and the extensive agricultural practice of utiliza tion of hybrid vigor on the other. Such utilization is the result of the economic value of hybrid combinations displaying superior yields and qualities as well as stability of performance, of benefits derived in breeding programs, and of the enhanced varietal protection of proprietary rights. No comprehensive and critical analysis of the phenomenon of heterosis in economic plants has been published for the last three decades since the now classical book Heterosis, edited by J . W. Gowen (Iowa State College Press, Ames, Iowa, 1952). MULCAHyl Pollen was long believed to serve primarily a single function, that of delivering male gametes to the egge A secondary and generally overlooked value of pollen is that it serves to block the transmission of many defective alleles and gene combinations into the next generation. This latter function comes about simply because pollen tubes carrying defective haploid genotypes frequently fail to complete growth through the entire length of the style. However, the beneficial consequences of this pollen selection are diluted by the fact that the same deleterious genotypes are often transmitted through the egg at strictly mendelian frequencies (Khush, 1973). Gene expression in the pollen might thus at least appear to be a phenomenon of trivial consequence. Indeed, Heslop-Harrison (1979) rightly termed the gametophytic portion of the angiosperm life cycle, the "forgotten generation." This neglect, however, came about despite subtle but constant indications that pollen is the site of intense gene activity and selection. For example, Mok and Peloquin (1975) demonstrated that relatively heterozygous diploid pollen shows heterotic characteristics whereas relatively homozygous diploid pOllen does not. Many individual aspects of the dynamics and assembly of biological membranes have been studied in great detail. Cell biological approaches, advanced genetics, biophysics and biochemistry have greatly contributed to an increase in our knowledge in this field.lt is obvious however, that the three major membrane constituents - lipids, proteins and carbohydrates- are studied, in most cases separately and that a coherent overview of the various aspects of membrane biogenesis is not readily available. The NATO Advanced Study Institute on "New Perspectives in the Dynamics of Assembly of Biomembranes" intended to provide such an overview: it was set up to teach students and specialists the achievements obtained in the various research areas and to try and integrate the numerous aspects of membrane assembly into a coherent framework. The articles in here reflect this. Statting with detailed contributions on phospholipid structure, dynamics, organization and biogenesis, an up to date overview of the basic, lipidic backbone of biomembranes is given. Extensive progress is made in the research on membrane protein biosynthesis. In particular the post- and co-translational modification processes of proteins, the mechanisms of protein translocation and the sorting mechanisms which are necessary to direct proteins to their final, intra - or extracellular destination have been characterized in detail. Modern genetic approaches were indispensable in this research area: gene cloning, hybrid protein construction, site directed mutagenesis and sequencing techniques elucidated many functional aspects of specific nucleic acid and amino acid sequences.
