In early 1973, I returned to Israel from a post-doctoral fellowship at Harvard University, and was accepted as a lecturer in the Department of Applied Microbiology at the Hebrew University of Jerusalem. Shortly after my return, Professor Richard Mateles, who at that time was head of the Department, suggested that I purchase a good and comprehensive book on single cell protein (SCP) in order to expand my general knowledge in the subject I had started then to work on; that was microbial utilization of one-carbon (C ) compounds. l Naturally, I took his advice (after all, he was the Boss) and bought the book, which was the only general book published on this subject at that time, and was based on papers presented at the First International Conference on Single Cell Protein, held at the Massachussetts Institute of Technology (M.I.T.), on October 1967 (Mateles and Tannenbaum, editors) [1]. Through this book I became acquainted with the world's hunger problem that existed in the past, and ways in which it was to be solved by SCP products prepared from CO , fossil-based raw 2 materials, and from wastes.

Measurements of different molecular species from single cells have the potential to reveal cell-to-cell variations, which are precluded by population-based measurements. An increasing percentage of researches have been focused on proteins, for its central roles in biological processes. Immunofluorescence (IF) has been a well-established protein analysis platform. To gain comprehensive insights into cell biology and diagnostic pathology, a crucial direction would be to increase the multiplexity of current single cell protein analysis technologies. An azide-based chemical cleavable linker has been introduced to design and synthesis novel fluorescent probes. These probes allow cyclic immunofluorescence staining which leads to the feasibility of highly multiplexed single cell in situ protein profiling. These highly multiplexed imaging-based platforms have the potential to quantify more than 100 protein targets in cultured cells and more than 50 protein targets in single cells in tissues. This approach has been successfully applied in formalin-fixed paraffin-embedded (FFPE) brain tissues. Multiplexed protein expression level results reveal neuronal heterogeneity in the human hippocampus.

SADRI, BEHROKH BAGHERIFAR. Protein Analysis at the Single Cell Level by Nonlinear Laser Wave-Mixing Spectroscopy for High Throughput Capillary Electrophoresis Applications. (Under the direction of William G. Tong and Morteza G. Khaledi) Nonlinear degenerate four-wave mixing is presented as an ultrasensitive optical absorption-based method for detection and measurement of biological samples. Wave-mixing imaging detection technique can localize and quantify biomolecules in single cells and tissue sections with excellent spatial distribution of light absorbed by a target sample. Cellular components can be label-free or labeled with a chromophore or a fluorophore and imaged by wave mixing using a CCD camera. In a 2-D forward-scattering wave-mixing geometry, two overlapping laser beams form interference gratings and transfer their energy to an absorbing medium, creating thermal gratings followed by changes in the refractive index. The probe beam diffracts off these laser- induced gratings to produce the signal beam, which is detected by a CCD camera or a photodiode. A single bio cell can be placed in a glass slide and as the laser beams probe the labeled cellular component, the CCD camera captures wave-mixing signals corresponding to the absorbing cellular components. This nonlinear imaging technique can be used for both live and fixed cells in real time to obtain information on sequential changes in the number, morphology and distribution of cellular components in a single cell. Nonlinear laser wave-mixing spectroscopy coupled with capillary electrophoresis provides a novel ultrasensitive method for single-cell protein analysis. This method is used to detect proteins separated within a single cell. Nonlinear wave mixing has many advantages including quadratic dependency on analyte concentration, high spatial resolution and small sample requirements.
