This book discusses the recent advancements in the role of various biomolecules in regulating root growth and development. Rhizobiology is a dynamic sub discipline of plant science which collates investigations from various aspects like physiology, biochemistry, genetic analysis and plant–microbe interactions. The physiology and molecular mechanisms of root development have undergone significant advancements in the last couple of decades. Apart from the already known conventional phytohormones (IAA, GA, cytokinin, ethylene and ABA), certain novel biomolecules have been considered as potential growth regulators or hormones regulating plant growth and development. Root phenotyping and plasticity analysis with respect to the specific functional mutants of each biomolecule shall provide substantial information on the molecular pathways of root signaling. Special emphasis provides insights on the tolerance and modulatory mechanisms of root physiology in response to light burst, ROS generation, agravitrophic response, abiotic stress and biotic interactions. In the last decade, enormous progress has been made on the physiology of plant roots, including on a wide range of molecular aspects. Much of that progress has been captured in the chapters of this book. Breakthroughs have been made possible through integration of molecular and whole-plant aspects. The classical boundaries between physiology, biochemistry and molecular biology have vanished. There has been a strong focus on a limited number of model species, including Arabidopsis thaliana. That focus has allowed greater insight into the significance of specific genes for plant development and functioning. However, many species are very different from A. thaliana, in that they are mycorrhizal, develop a symbiosis with N2-fixing microsymbionts, or have other specialised root structures. Also, some have a much greater capacity to resist extreme environments, such as soil acidity, salinity, flooding or heavy-metal toxicities, due to specific adaptations. Research on species other than A. thaliana is therefore pivotal, to develop new knowledge in plant sciences in a comprehensive manner. This fundamental new knowledge can be the basis for important applications in, e.g., agriculture and plant conservation. Although significant progress has been made, much remains to be learnt. It is envisaged that discoveries made in the recent past will likely lead to major breakthroughs in the next decade.

In 1971, the late Dr. J. Kolek of the Institute of Botany, Bratislava, organized the first International Symposium devoted exclusively to plant roots. At that time, perhaps only a few of the participants, gathered together in Tatranska Lomnica, sensed that a new era of root meetings was beginning. Nevertheless, it is now clear that Dr. Kolek's action, undertaken with his characteristic enormous enthusiasm, was rather pioneering, for it started a series a similar meetings. Moreover, what was rather exceptional at the time was the fact that the meeting was devoted to the functioning of just a single organ, the root. One possible reason for the unexpected success of the original, perhaps naive, idea of a Root Symposium might lie with the fact that plant roots have always been extremely popular as experimental material for cytologists, biochemists and physiologists whishing to probe processes as diverse as cell division and solute transport. Of course, the connection of roots with the rest of the plant is not forgotten either. This wide variety of disciplines is now coupled with the development of increasingly sophisticated experimental techniques to study some of these old problems. These factors undoubtedly contribute to the necessity of continuing the tradition of the root symposia. The common theme of root function gives, in addition, a certain unity to all these diverse activities.

Although the role of salicylic acid (SA) in plant physiological processes has been widely studied for a long time, many open questions remain several fields. The importance of SA synthesis is illustrated by the four review papers published in this Special Issue that represent a wide range of approaches, indicating that a growing body of evidence needs to be summarized in a thought-provoking manner. The investigations presented in the six original studies extend upon the understanding of the involvement of SA in anthracnose infection and light-dependent cold acclimation, highlighting the use of SA mutant Arabidopsis plants. The studies also focused on the application of novel SA analogs or SA in combination with Rhizobacteria inoculation. We hope that the four reviews and six studies provide a deeper understanding of the role of SA and its complex tasks, as well as a new direction for research to address gaps and open questions, including both at the metabolite and gene expression levels, in the use of agriculturally important crop or mutant model plants, and in both basic research and practical applications.

The appearance of the new generation in higher plants is ensured by the presence of viable seeds in the mother plant. A good number of signaling networks is necessary to provoke germination. Phytohormones play a key role in all stages of seed development, maturation, and dormancy acquisition. The dormancy of some seeds can be relieved through a tightly regulated process called after-ripening (AR) that occurs in viable seeds stored in a dry environment. Although ABA is directly involved in dormancy, recent data suggest that auxin also plays a preponderant role. On the other hand, the participation of reactive oxygen species (ROS) in the life of the seed is becoming increasingly confirmed. ROS accumulate at different stages of the seed’s life and are correlated with a low degree of dormancy. Thus, ROS increase upon AR and dormancy release. In the last decade, the advances in the knowledge of seed life have been noteworthy. In this Special Issue, those processes regulated by DOG1, auxin, and nucleic acid modifications are updated. Likewise, new data on the effect of alternating temperatures (AT) on dormancy release are here present. On the one hand, the transcriptome patterns stimulated at AT that encompasses ethylene and ROS signaling and metabolism together with ABA degradation were also discussed. Finally, it was also suggested that changes in endogenous γ-aminobutyric acid (GABA) may prevent seed germination.
