Populations of cells were obtained from a clinic offering "treatments" for chronic spinal cord injury. Immunostaining and flow cytometry confirmed the populations to be neural and mesenchymal stem cells, respectively (NSCs and MSCs). ELISAs performed on conditioned media detected several growth factors although neurotrophic factors known to promote regeneration in the spinal cord were not detected. Treatment of primary cortical neurons with media conditioned by MSCs and NSCs did not lead to a statistically significant increase in axon length or complexity. A statistically significant increase in cell number was detected in cultures treated with NSC-conditioned media. In vitro analysis does not support the clinic's claim that the cells exhibit properties which would promote axon growth. Two human embryonic stem cell lines (ES) and three induced pluripotent stem cell lines (iPS) were differentiated into high purity populations of neuronal progenitor cells (NPCs) and oligodendrocyte progenitor cells (OPCs). Both ES and two of three iPS lines produced high purity populations of the desired lineage. Transplantation of OPCs generated from ES and iPS lines into spinal cord injured rats resulted in engraftment of human oligodendrocytes without formation of tumors. We conclude that iPS cells may be adapted to protocols developed for generation of high purity NPCs and OPCs from ES cells. Immature astrocytes are known to promote recovery from spinal cord injury. In order to determine properties of immature astrocytes which promote recovery after spinal cord injury an in vitro model of aged astrocytes was developed. As compared to aged astrocytes, a greater proportion of immature astrocytes are proliferating, express more laminin, are motile, and promote greater regeneration of dorsal root ganglion (DRG) axons. Reprogramming mature astrocytes into immature astrocytes is a potential therapy for chronic spinal cord injury. Nine factors which are expressed at higher levels in immature versus mature astrocytes were overexpressed in aged astrocytes. At two weeks, treated cultures exhibited greater laminin immunofluorescence, increased capacity to promote growth of adult DRG axons, and foci of increased motility mixed with foci of cell death. Treatment of aged astrocytes with 9 factors leads to partial recovery of immature properties.
