Prostaglandin Research reviews the status of prostaglandin research, with particular emphasis on chemistry. This book highlights the role of scientific research in society in attempting to find drugs, with potential application to therapy, the chief impetus for prostaglandin research. This volume is organized into seven chapters and begins with an overview of the origin and properties of prostaglandins and then outlines the stage of research in the area of biosynthesis of natural entities, especially endoperoxides and thromboxanes. The reader is then introduced to a rich source of natural prostaglandins, the gorgonian Plexaura homomalla, and the pharmacology of prostaglandins. The physical methods used in prostaglandin research are considered, along with the chemistry of natural and modified prostaglandins. This book is aimed at experts in the field, as well as scientists with a general interest in chemistry and biology.

This is the second in a series of three books on advances in prostaglandin research. In recent years there has been an unparalleled interest in these compounds and as a result a vast amount of research data has accumulated since the publication of my earlier book in 1972. At that time it was possible to present a fairly comprehensive review of the various aspects of prosta glandin research in one volume. This is no longer possible and the contents are divided into three volumes; the first one dealing with prostaglandins and reproduction was published in October 1975; the present volume dealing with chemical and biochemical aspects of prostaglandin research, to be published in April 1976 and the third book dealing with physiological, pharmacological and pathological aspects of prostaglandin research wiII be published in May 1976. The authorship represents international scientists consisting of physiolo-. gists, pharmacologists, chemists, biochemists, veterinary scientists and obstetrician gynaecologists actively engaged in different areas of prosta glandin research. An attempt has been made to provide a total coverage of advances relating to prostaglandins. For the sake of completeness and continuity, material covered in the 1972 book is either briefly summarised or reference made to that edition. In recent years there have been notable advances in the chemical and biochemical aspects of prostaglandin research and these are discussed by various authorities in the chapters that follow.

The International Symposium on Prostaglandins and Related Compounds, first held in Vienna 1972, revisited the city after 24 years for the 10TH Symposium. For the many re searchers working in this multi-disciplinary field it was an opportunity to exchange their ex periences and share new data with colleagues from all around the world. This scientific exchange was largely encouraged by the unseasonably cold and rainy weather. For the first time, there was quite a large attendance from the former Communist countries. Eugene Garfield prepared a key note address delivered during the meeting (The Sci entist 1996, 12) reviewing the contribution of the Nobel Laureates U.S. von Euler, l.R. Vane, S.K. Bergstrom, and B.I. Samuelsson, discussing the relevance of the more than 40,000 pa pers in this area published since 1991. Overall, there is still a rapidly growing interest, and in particular a great variety of clinical applications of this family of compounds which were dis cussed in detail during the meeting. Beside the lectures there were 19 workshops covering nearly all the topics of key in terest. All the speakers were invited to prepare a manuscript which has resulted in the volume now in your hands. Special thanks to Dr. Patrick Wong and the new publisher of this series who helped to publish the proceedings in the usual quality and reasonable period of time. Looking forward to seeing all of you again in Florence in 1999, hopefully with much more sun.

The voluminous literature on prostaglandins includes some excellent reviews. For some time however, there has been a need for the collection in one volume of information from individual experts actively engaged in different aspects of prostaglandin research. This book is an attempt to fulfil that need. In view of the expanding interest and ever-growing accumulation of data of the various effects of prostaglandins it is hoped to revise the book and bring it up-to-date periodically. The largest section is that on reproduction, in which field progress in research and clinical applications thereof have been, and are, of great significance particularly in the area of popula tion control. This is not to decry the skill and efforts of investi gators in other aspects of prostaglandin activity nor the potential biological or therapeutic importance of their findings. In this context, what may appear to be omissions are individual sections on prostaglandin activity in the central nervous system, in the eye and as mediators of the inflammatory response. These are considered in the chapter on General Pharmacology of the Prostaglandins not because they might be considered to be of lesser impact and significance but in order to achieve a rapid publication of the book. This volume is the proceedings of the 11th International Conference on Advances in Prostaglandin and Leukotriene Research: Basic Science and New Clinical Applications, held in Florence, Italy, on June 4-8, 2000, which continues the tradition of covering both basic and clinical aspects of prostaglandin and leukotriene research. At this meeting particular emphasis was given to the potential application of the novel COX-2 inhibitors, the genetics of asthma relating to the production of leukotrienes, and novel cellular networks for the production of leukotrienes and lipoxins. Traditionally, as has been done in past meetings of this series, a considerable amount of new material was presented and discussed this year, with particular attention given to the newest clinical data.

Prostaglandin research is one of the most explosive fields in the biological science, and there have been a number of international symposia on this subject. In planning the second Advanced Course in Erice (Sicily) at the "Ettore Majorana" center it was realized that large congresses might not represent the best way to facilitate communication within specialized areas of biologic and medical research. In fact small meetings where in depth discussion is confined to a limited number of scientists, are now generally considered successful vehicles of scientific information. With this in mind, we made an effort in collecting a number of experts in both experimental and clinical use of these important arachidonate metabolites. We have been very fortunate in bringing together a faculty of international authorities who contributed to the course with brilliant presentations and lively discussion. Thus, this book represents an up-to-date overview of the physiology and pharmacology of different products derived from arachidonic acid metabolism, associated with outstanding suggestions of their clinical potential. It is our hope that from this Advanced Course, and its resulting publication, new ideas concerning the physio pathological role of these compounds will arise together with new directions for future research. We are deeply indebted to NATO for its interest and generous support which made this Second Course a reality.

This is the fourth in a series of books on Advances in Prostaglandin Research published by MTP Press. From laboratory and animal studies a large number of practical applications of prostaglandins have been identified. In some areas clinical trials have also been carried out. There is usually a long time interval between identifying practical applications of new drugs and clinical trials. Most of the information on practical applications of prostaglandins is scattered in different journals. Although a number of books dealing with prostaglandin research have been published (including some on practical applications in specific areas, e.g. reproduction) there is no single volume dealing with all areas of practical applications of these substances. This book aims at filling this gap and should be of interest to clinicians as well as to medical and basic scientists in academic institutions and in the pharmaceutical industry. In some areas, exciting potential applications of prostaglandins have been identified from basic research but clinical trials have so far not been carried out. Such areas of practical applications are covered in a separate intro ductory chapter. All contributors to the volume are scientists and clinicians actively engaged in prostaglandin research and most are recognized authorities in specific areas. The need for rapid publication in a fast-expanding field is obvious.

Arachidonic acid metabolites are known to playa regulatory role in a number of biological systems, in which they function as microenviron mental hormones and intracellular signal mediators. One of the most exciting areas of research of these compounds is the one that studies the relationship between prostaglandins and tumor cell growth and function. In the last few years there has been an extraordinary evolution of data on prostaglandins (and related compounds) and cancer. This vol ume is based on papers presented at the 1986 International Confer ence on Prostaglandins and Cancer organized by the Italian National Research Council and the II University of Rome, and held in Rome, Italy, in June, 1986. This Conference brought together oncologists and specialists in the areas of prostaglandin chemistry, biochemistry, pharmacology, physiology, cellular and molecular biology to overview the actual state of knowledge on the role of eicosanoids in cancer and to focus on the key questions that need to be answered. The picture that comes out of this book describes a very complicated network of interactions between arachidonic acid metabolites and different as pects of the complex phenomenon "cancer". Eicosanoids participate in carcinogenesis initiation and promotion, and their relationship with tumor promoters and growth factors is well established. During cancer growth, different prostaglandins can have different roles in the regulation of cell proliferation and differentiation and in metastasis formation; meanwhile evidence is accumulating for a pos sible use of some of these compounds as antineoplastic agents.

Prostaglandins are hormone-like substances that participate in a wide range of body functions such as the contraction and relaxation of smooth muscle, the dilation and constriction of blood vessels, control of blood pressure, and modulation of inflammation. Prostaglandins are derived from a chemical called arachidonic acid. This book presents the latest research in this field.
