Research on purine derivatives as potent and selective modulators of physiologic functions has moved to center stage. This volume covers the biology and chemistry of purines and of their receptors broadly under these section headings: - Adenosine Receptors and Effector Systems - Adenine Nucleotide Receptors and Effector Systems - Phosphodiesterase Enzymes and Inhibitors The presentations focus on the potential development of clinically useful drugs and powerful agents that activate or block purinergic receptors or that inhibit phosphodiesterases. Xanthines (caffeine and theophylline) represent one class of such agents. This volume is for pharmacologists, biochemists, and medical chemists in research labs of universities, gov- ernment, and the pharmaceutical industry.

Purine receptors are a type of plasma membrane molecules found in nearly all mammalian tissues. They are classified into three categories and include P1 receptors, P2X receptors, and P2Y receptors. They all play a key role in the variety of neuronal and non-neuronal mechanisms, which comprise pain, immune responses, exocrine and endocrine secretion, platelet aggregation, endothelial-mediated vasodilatation, and inflammation. The G protein is mainly bound to the P1 and P2Y receptors. These receptors are differentiated by considering their reactivity towards particular activators. The brain, heart, kidneys, and adipose tissue all contain P1 and P2Y receptors. These receptors are responsible for various cellular functions, comprising vascular reactivity, migration and proliferation of neural stem cells, cytokine secretion and apoptosis. The study of purine receptors is helpful for developing therapeutic strategies for those disorders, which have new action mechanisms, such as pathogenic conditions, like trauma, pain, epilepsy, ischemia, drug dependence and psychiatric disorders. The objective of this book is to present advanced studies on purine receptors as well as their pharmacological applications. Those in search of information to further their knowledge will be greatly assisted by it.

Physiological, pharmacological and molecular biological data generated over the past three decades have demonstrated the existence of two major families of extracellular receptors, the P1, a family of four G-protein coupled receptors and the P2, a family of at least 12 receptors responsive to purine (ATP, ADP) and pyrimidine (UTP) nucleotides through which adenosine and ATP can function as extracellular messengers. The present two-part volume represents an integrated compendium of invited chapters by leading researchers in the area focusing on advances in the understanding of purinergic and pyrimidinergic signaling systems, their role(s) in tissue function and pathophysiology and advances in developing potential new medications based on the modulation of P1 and P2 receptor signaling processes. The volumes will thus provide the reader with a topical, comprehensive and integrated overview of this important area.

This book aims to provide a brief update on the functions of purinergic receptors in various systems, in addition to the signaling pathway activated to mediate these functions. We address the influence of hypoxia by modulating the activity of these receptors under physiological and pathophysiological conditions. Additionally, we describe the mechanisms of induction of pain and inflammation in different systems. Finally, the book discusses some of the main bioinformatics tools currently used to improve or discover new prototypes capable of selectively acting on these receptors with estimated parameters of satisfactory solubility and toxicity for possible commercial implementation.

Edited by Kenneth A. Jacobson and Michael F. Jarvis The roles of extracellular purines and pyrimidines in cellularhomeostasis and disease etiology have come to be understoodgradually over the past 40 years. However, due to the recentcloning and expression of receptors for ATP and adenosine, novelcompounds have been developed with unique therapeutic potential forthe treatment of thrombosis, stroke, epilepsy, chronic pain, immunological disorders, and cancer. As a result, the study ofadenosine- and ATP-mediated responses in cellular regulation isentering a phase of opportunity and development unmatched since thedays of serotonin receptor research in the 1970s. The only definitive book on the topic, Purinergic Approaches inExperimental Therapeutics covers all of the major therapeuticapplications of purinergic receptors and reflects the very latestdevelopments in this new area of therapeutic research. Twenty-eightchapters, authored by an international group of contributors whoare the leading authorities in the field, provide details onmolecular pharmacology; medicinal chemistry; and therapeuticimplications, including cardiology, metabolism, immunology, neurology, and cancer. Among the topics covered: * Purinergic Neurotransmission and Neuromodulation: A HistoricalPerspective * Adenosine Receptor Subtypes: New Insights from Cloning andFunctional Studies * Modulators of Adenosine Uptake, Release, and Inactivation * Cardiac Electrophysiology of Adenosine: Antiarrhythmic andProarrhythmic Actions * Purinergic Modulation of Gastrointestinal Function * The Role of Adenosine in Asthma * ATP in Brain Function * ATP in the Treatment of Cancer For researchers in pharmacology, physiology, molecular biology, andmedicinal chemistry, Purinergic Approaches in ExperimentalTherapeutics heralds an exciting new era in the understanding ofpurinergic neurotransmission and the development of noveltherapeutic modalities.
