Experiments have been done that compare the actions of botulinum toxin type A, tetanus toxin, diphtheria toxin and beta-bungarotoxin on the cholinergic neuromuscular junction. All four toxins bind to tissues without producing adverse effects; each of the toxins has its own unique class of receptors. After tissue binding has occurred, botulinum toxin, tetanus toxin and diphtheria toxin are internalized; this process is antagonized by chloroquine, ammonium chloride and methylamine hydrochloride. Beta-Bungarotoxin is not internalized and is not antagonized by the drugs just listed. Diphtheria toxin acts intracellularly to ADP-ribosylate elongation factor 2. The intracellular actions of botulinum toxin and tetanus toxin have not been determined.

In this first annual report we present our progress in three of the Statement of Work. In addition we have included our work being done in collaboration with Walter Reed Army - Institute of Research. Structural work with Clostridium botulinum B and a potential inhibitor, SABIM, is reported. Though it was predicted that it would bind to the active site, we have shown that it binds at two sites in BoNT/B, the active site and the substrate-binding site. We speculate that the inhibitory property of this molecule may be because it chelates the active site zinc or because it blocks the substrate binding site or due to both. We also propose that analogs of BABIM might work better. We are continuing with our studies on BoNT/B with other potential inhibitors. A set of potential inhibitors identified by biochemical methods and a few small molecules known to inhibit zinc - endopeptidases like thermolysin and carboxypeptidase have been screened by computational methods before determining the structures of the complexes. Combining the docking calculations with the x-ray diffraction methods offers a powerful rational design of inhibitors. From our studies so far we conclude that small molecules that will stay bound to the toxins will%be the best inhibitors.

In this second annual report we present our progress on three different areas. We are working on intact BoNT/B to identify small molecules that could be used to block the toxic activity. In this respect, we have discovered that two calcium ions are bound to BoNT/B and that at least one of them plays an important role in the translocation of the catalytic domain into the cytosol. We propose that this calcium ion site could be targeted by calcium specific chelators to block translocation. As for the ganglioside binding site as a target, we are working with both intact BoNT/B and structurally similar C fragment of tetanus neurotoxin. We have identified two molecules that might compete with ganglioside for binding which could potentially be used as inhibitor. One of our Statements Of Work is to work with Clostridium botulinum neurotoxin E. Since the diffraction quality of intact toxin is poor, it is difficult to use the intact toxin for inhibitor study via x-ray crystallography. We are using BoNT/E light chain to this inhibitor study and have determined the crystal structure of BoNT/E light.
