Molecular imaging is visualizations and measurements of in vivo biological processes at the molecular or cellular level using specific imaging probes. As an emerging technology, biocompatible macromolecular or nanoparticle based targeted imaging probes have gained increasing popularities. Those complexes consist of a carrier, an imaging reporter, and a targeting ligand. The active targeting ability dramatically increases the specificity. And the multivalency effect may further reduce the dose while providing a decent signal. In this thesis, sentinel lymph node (SLN) mapping and cancer imaging are two research topics. The focus is to develop molecular imaging probes with high specificity and sensitivity, for Positron Emission Tomography (PET) and optical imaging. The objective of this thesis is to explore dextran radiopharmaceuticals and porous silicon nanoparticles based molecular imaging agents. Dextran polymers are excellent carriers to deliver imaging reporters or therapeutic agents due to its well established safety profile and oligosaccharide conjugation chemistry. There is also a wide selection of dextran polymers with different lengths. On the other hand, Silicon nanoparticles represent another class of biodegradable materials for imaging and drug delivery. The success in fluorescence lifetime imaging and enhancements of the immune activation potency was briefly discussed. Chapter 1 begins with an overview on current molecular imaging techniques and imaging probes. Chapter 2 presents a near-IR dye conjugated probe, IRDye 800CW-tilmanocept. Fluorophore density was optimized to generate the maximum brightness. It was labeled with 68Ga and 99mTc and in vivo SLN mapping was successfully performed in different animals, such as mice, rabbits, dogs and pigs. With 99mTc labeled IRDye 800CW-tilmanocept, chapter 3 introduces a two-day imaging protocol with a hand-held imager. Chapter 4 proposed a method to dual radiolabel the IRDye 800CW-tilmanocept with both 68Ga and 99mTc. Chapter 5 introduces a 68Ga metal chelating bioorthogonal tetrazine dextran probe for multistep imaging of a colon cancer. Chapter 6 presents the synthesis and in vivo evaluation of a Hepatocellular Carcinoma targeting PET probe 68Ga-Insulin-Dextran. Chapter 7 discusses a novel method to prepare silicon nanoparticles with great yield and size control. The last chapter 8 concludes all probes developed in this thesis and their clinical relevance.

Positron emission tomography (PET) is the one molecular imaging modality that can be used in both pre-clinical and human studies. PET provides the most direct diagnoses in oncology with high sensitivity and quantitative analysis. The primary reason is that it can demonstrate dynamic metabolism in animal or human bodies during in vivo molecular imaging. However PET is not only confined to oncology. It can be used in other physiological or metabolic processes, as long as specific biomarkers are available. Therefore, the development of radiotracers is crucial for in vivo PET imaging. Synthetic chemistry provides a platform to synthesize cold standards (the counterpart of the non-radioactive material of PET radiotracers) and radio-labeling precursors. Once the compounds with high specificity and binding affinity are developed, chemical synthesis can make radiolabeling precursors for 11C, 18F or other positron emitting isotopes. In this thesis, two series of molecular probes are synthesized as PET imaging agents. One series of molecules are AP site specific binging agents and the other series of molecules are myelin specific probes. The former one is used for in vivo PET imaging of the AP site formation and can be used as imaging markers to evaluate DNA-targeted chemotherapeutic drugs. The latter type molecules are myelin-binding agents for in vivo imaging of myelination in the central nervous system (CNS). This thesis presents the application of synthetic chemistry in the development of 18F-labeled probes in two projects: development of [18F]-labeled PET tracers for imaging of DNA damage and repair in cancer chemotherapies and development of [18F]-labeled PET imaging probes for in vivo studies of myelin pathology

Nowadays, imaging techniques are an indispensable tool in cancer research, clinical trials and medical practice and they are of great aid for oncologists to localize and characterize abnormal tissues. Detection and characterization of lesions, especially tumors, can be achieved by using specific tracers or contrast media that are capable of selectively interact with the biological target. This approach is defined as Molecular Imaging. A MI probe is usually composed of a biologically active component (vector) that interacts with the intended molecular target, an imaging reporter that should produce a detectable signal and a linker between the two components. Among the different imaging modalities only a few are characterized by the merging of these functional units into one molecular identity: PET tracers and Hyperpolarized MRI probes use labeled nuclides (either radioactive or stable) as imaging reporters within the biologically active molecule. My work has dealt with the development of hyperpolarized MRI tracers, which are able to overcome the sensitivity issue typical of the MRI methodology avoiding the radiological concerns that negatively affect the use of nuclear techniques.
