Nonlinear chromatography is a field that borders both chemical engineering and physical chemistry. In turn, the theory of nonlinear chromatography is the foundation of preparative chromatography, a separation process that has lately become of considerable interest in the pharmaceutical industry. Only chromatography is sufficiently flexible and powerful to satisfy the practical requirements encountered in most difficult separations of pharmaceuticals and pharmaceutical intermediates. Since "nonlinear" behaviour is strictly a mathematical concept, it is difficult to leave mathematics out of any fundamental study of nonlinear chromatography. Therefore, Modeling for Preparative Chromatography will describe the different mathematical models of chromatography, examine the assumptions on which they are based, consider their properties, and discuss their solutions. All this will be done from a mathematical analysis point of view, paying considerable attention to the influence of nonlinear behavior on the results. Clear and simple discussions of the basic physico-chemical phenomena involved in HPLC Clear and complete presentation of the relevant properties of the mathematical tools involved Detailed analysis of nonlinear effects

Completely revised and substantially extended to reflect the developments in this fast-changing field. It retains the interdisciplinary approach that elegantly combines the chemistry and engineering involved to describe the conception and improvement of chromatographic processes. It also covers recent advances in preparative chromatographic processes for the separation of "smaller" molecules using standard laboratory equipment as well as the detailed conception of industrial chemical plants. The increase in biopharmaceutical substances is reflected by new and revised chapters on different modifications of continuous chromatography as well as ion-exchange chromatography and other separation principles widely used in biochromatography. Following an introductory section on the history of chromatography, the current state of research and the design of chromatographic processes, the book goes on to define the general terminology. There then follow sections on stationary phases, selection of chromatographic systems and process concepts. A completely new chapter deals with engineering and operation of chromatographic equipment. Final chapters on modeling and determination of model parameters as well as model based design, optimization and control of preparative chromatographic processes allow for optimal selection of chromatographic processes. Essential for chemists and chemical engineers in the chemical, pharmaceutical, and food industries.

Download Fundamentals of Preparative and Nonlinear Chromatography Book in PDF, ePub and Kindle
The second edition of Fundamentals of Preparative and Nonlinear Chromatography is devoted to the fundamentals of a new process of purification or extraction of chemicals or proteins widely used in the pharmaceutical industry and in preparative chromatography. This process permits the preparation of extremely pure compounds satisfying the requests of the US Food and Drug Administration. The book describes the fundamentals of thermodynamics, mass transfer kinetics, and flow through porous media that are relevant to chromatography. It presents the models used in chromatography and their solutions, discusses the applications made, describes the different processes used, their numerous applications, and the methods of optimization of the experimental conditions of this process.

Chromatographic separations are usually realized in isocratic mode, i.e., with constant temperature, pressure or solvent composition of the mobile phase during the process. However, it is well known that the adsorption behavior of the system can be strongly affected by changes of these operating parameters in gradient mode. In the work presented here several aspects of applying the gradient technique for preparative scale of both batch and continuous chromatography are discussed. This analysis is included in the main PART II of the work. It starts from the examination of a batch system consisting of a single column. Subsequently, strategies of using gradient in a continuous separation with a parallel multi-column arrangement and with the SMB system are discussed. Before analyzing the gradient technique the application of mathematical models of process dynamics for predicting chromatographic separations is thoroughly examined in PART I of the work. Moreover, in PART I several aspects of the application of numerical methods for solving the model equations are discussed.
