The object of this thesis research was to develop and characterize a new type of acoustic biosensor - a ZnO-based laterally excited thickness shear mode (TSM) resonator in a solidly mounted configuration. The first specific aim of the research was to develop the theoretical underpinnings of the acoustic wave propagation in ZnO. Theoretical calculations were carried out by solving the piezoelectrically stiffened Christoffel equation to elucidate the acoustic modes that are excited through lateral excitation of a ZnO stack. A finite element model was developed to confirm the calculations and investigate the electric field orientation and density for various electrode configurations. A proof of concept study was also carried out using a Quartz Crystal Microbalance device to investigate the application of thickness shear mode resonators to cancer biomarker detection in complex media. The results helped to provide a firm foundation for the design of new gravimetric sensors with enhanced capabilities. The second specific aim was to design and fabricate arrays of multiple laterally excited TSM devices and fully characterize their electrical properties. The solidly mounted resonator configuration was developed for the ZnO-based devices through theoretical calculations and experimentation. A functional mirror comprised of W and SiO2 was implemented in development of the TSM resonators. The devices were fabricated and tested for values of interest such as Q, and electromechanical coupling (K2) as well as their ability to operate in liquids.

ABSTRACT: Acoustic wave sensors have proven useful in many fields as primarily mass sensitive devices capable of responding to small environmental perturbations. The focus of this dissertation is the development of a new type of surface acoustic wave device with application to material property measurement, and biological and chemical sensing. This device is a combination of three independent acoustic wave devices with these waves propagated across the same area, while retaining independence of actuation and sensor function. The development of a complete sensor system, and its use and operation are presented for several example cases of chemical and biomarker sensing, and sample manipulation. These include experimental and theoretical studies for organic vapor sensing, biological moiety sensing, acoustic streaming to remove loosely bound material, and optimization of designs for these applications.

There has been a renewed interest in the development of surface acoustic wave (SAW) biosensors because they hold great promise for opening new frontiers in biology and medicine. The promise of SAW technology is grounded in the advantages SAW devices hold over traditional laboratory techniques used in biological and medical laboratories. These advantages include having smaller sizes to allow greater portability, using smaller sample volumes, requiring lower power requirements, the ability to integrate them into microfluidic platforms, and their compatibility with smart devices such as smartphones. The devices offer high sensitivity and can be designed to allow microfluidic interfacing. Other major advantages of SAW-based technologies include the fact that they can be operated remotely in harsh conditions without the need for an AC power supply. Their compatibility with lab-on-a-chip systems allows the creation of fully integrated devices with the ability to isolate the sample from the operator. In this mini-review, we will discuss SAW devices and their ability to enable a variety of applications in Biology and Medicine. The operating principles of the SAW biosensors will be discussed along with some technological trends and developments.

The object of this thesis research is to facilitate the appraisal and analysis of the signatures of the modern acoustic wave biosensors, as well as to improve the experimental methodology to enhance sensor performance. For this purpose, both theoretical characterization of acoustic wave sensor signatures and experimental studies for the most frequently used acoustic wave biosensors, the liquid phase QCM (quartz crystal microbalance) and the vapor phase SAW (surface acoustic wave) sensors, are presented. For the study of SAW vapor phase detection, the author fabricated different types of two-port SAW resonator sensors on quartz substrates and designed and performed a significant number of detection experiments. These were conducted both with calibrated or known target samples under laboratory conditions at Georgia Tech Hunt Lab and with samples of unknown concentrations such as seized crack cocaine (courtesy of Georgia Bureau of Investigation, GBI) to see the sensors capability to work in the field conditions. In addition, the dependence of the SAW sensor signatures on specific locations of the surface perturbation was investigated to account for some observed abnormal responses. Finally, a novel approach to classify and visualize chemically analogous substances is introduced. The author expects that the thesis work herein may contribute to the study of the modern acoustic wave biosensors which includes but is not limited to: the establishment of underpinning theory that will aid in the evaluation of the signatures; the practical aspects of design and fabrication of SAW devices specific to the vapor phase immunoassay; the development of efficient experimental methodologies; the strategic immobilization of a biolayer on SAW resonator based biosensors; and, the acquisition of reference data for the development of commercial acoustic wave sensors.
