There has been an increasing interest in development of thin film bulk acoustic wave resonator devices for chemical and biological sensing applications in the environmental and biomedical industries. The zinc oxide thin film FBAR devices provide attractive advantages, such as high resonant frequency, small size, and rapid response. Typically, the ZnO-based FBAR devices are operated in the longitudinal wave mode where the ZnO crystallites are perpendicular to the substrate. However, the longitudinal wave mode is adversely affected when used in liquid environments. To overcome this limitation, the shear mode solidly mounted film bulk acoustic wave resonator device is presented in this dissertation.

The analysis and development of robust sensing platforms based on solidly-mounted ZnO bulk acoustic wave devices was proposed. The exploitation of acoustic energy trapping was investigated and demonstrated as a method to define active sensing areas on a substrate. In addition, a new "hybrid" acoustic mode experiencing acoustic energy trapping was studied theoretically and experimentally. This mode was used as an explanation of historical inconsistencies in observed thickness-shear mode velocities. Initial theoretical and experimental results suggest that this mode is a coupling of thickness-shear and longitudinal particle displacements and, as such, may offer more mechanical and/or structural information about a sample under test. Device development was taken another step further and multi-mode ZnO resonators operating in the thickness-shear, hybrid, and longitudinal modes were introduced. These devices were characterized with respect to sample viscosity and conductivity and preliminary results show that, with further development, the multi-mode resonators provide significantly more information about a sample than their single-mode counterparts. An alternative to resonator-based platforms was also presented in the form of bulk acoustic delay lines. Initial conceptual and simulation results show that these devices provide a different perspective of typical sensing modalities by using properly designed input pulses, device tuning, and examining overall input and output signal spectra.

In this research, I present several novel contributions to the field of microelectronic acoustic biosensors that approach the goal of developing a cost-effective, highly sensitive and highly selective sensor array for the detection of early cancer proliferation. First I report on the development of a novel solidly mounted shear-mode resonator employing piezoelectric ZnO as an appropriate base device for liquid-phase sensing applications. Second I report on the development of a novel and appropriate chemical surface preparation protocol for the covalent immobilization of monoclonal IgG antibodies to the surface of this device, thereby functionalizing it as a biosensor. Proof of functionality is demonstrated employing these sensors for the detection of various known cancer biomarkers in complex media as well as progress towards developing a packaged disposable sensor array system is also reported.

This thesis investigates the usage of surface acoustic wave (SAW) devices as sensors in the determination of viscosity of Newtonian fluids such as Bovine Serum Albumin (BSA). Specifically, conductance and susceptance data of the SAW device in the presence of Bovine Serum Albumin and other liquids, comprising of proteins in small quantities (~35 nL). A two-port SAW device (320 MHz) was fabricated and tested with different concentrations of a water-miscible BSA in deionized water (DI water) and glycerol. The study examines several designs and type of substrates. There is a strong correlation of the measured sensor data to the corresponding computer simulations. A Finite Element Model (FEM) of the Two-Port Surface Acoustic Device is used to simulate the Sensor characteristics. COMSOL Multiphysics Modeling and simulation of surface acoustic wave devices are also utilized for protein characterization. The models predict how the presence of fluid in the groove lowers the resonance frequency. The SAW Sensor simulation detects changes in the viscosity of a protein using thermoviscous acoustic - piezoelectric interactions. The model accurately predicts the behavior of the SAW and fluid interactions. Rigorous analysis of viscosity and resultant variations in conductance, susceptance, and admittance, as a function of Protein concentration and morphology, establish SAW sensors as significant characterization devices. New drug discovery and development is costly; of all compounds investigated for use in humans, only small fractions are approved for treatments. The pharmaceutical industry has started switching from molecular drugs to protein-based drugs due to high specificity, enhanced efficacy, affinity, solubility, and low toxicity. Novel proteins in elevated concentrations are very expensive to produce. On the other hand, their solution properties, especially the viscosity is very critical to the stability, processing, manufacturing, and delivery of such drugs.

Acoustics is a mature field which enjoys a never ending youth. New developments are induced by either the search for a better understanding, or by technological innovations. Micro-fabrication techniques introduced a whole new class of microdevices, which exploit acoustic waves for various tasks, and in particular for information processing and for sensing purposes. Performance improvements are achievable by better modelling tools, able to deal with more complex configurations, and by more refined techniques of fabrication and of integration in technological systems, like wireless communications. Several chapters of this book deal with modelling and fabrication techniques for microdevices, including unconventional phenomena and configurations. But this is far from exhausting the research lines in acoustics. Theoretical analyses and modelling techniques are presented, for phenomena ranging from the detection of cracks to the acoustics of the oceans. Measurement methods are also discussed, which probe by acoustic waves the properties of widely different systems.
