Lignin, a complex aromatic polymer in terrestrial plants, contributes significantly to biomass recalcitrance to microbial and/or enzymatic deconstruction. To reduce biomass recalcitrance, substantial endeavors have been exerted on pretreatment and lignin engineering in the past few decades. Lignin removal and/or alteration of lignin structure have been shown to result in reduced biomass recalcitrance with improved cell wall digestibility. While high lignin content is usually a barrier to a cost-efficient application of bioresources to biofuels, the direct correlation of lignin structure and its concomitant properties with biomass remains unclear due to the complexity of cell wall and lignin structure. Advancement in application of biorefinery to production of biofuels, chemicals, and bio-derived materials necessitates a fundamental understanding of the relationship of lignin structure and biomass recalcitrance. In this mini-review, we focus on recent investigations on the influence of lignin chemical properties on bioprocessability--pretreatment and enzymatic hydrolysis of biomass. Furthermore, lignin-enzyme interactions and the effects of lignin compositional units, hydroxycinnamates, and lignin functional groups on biomass recalcitrance have been highlighted, which will be useful not only in addressing biomass recalcitrance but also in deploying renewable lignocelluloses efficiently.

Due to its depletion and the environmental damage it causes, hydrocarbons are being replaced by energy from renewable sources. One such form of energy source is Biomass. Biomass is a renewable raw material generated by living organisms and found in agricultural waste in large quantities. The three main components of biomass are cellulose, hemicellulose and lignin. The first two components are sugar polymers, being cellulosic ethanol a desirable goal for converting those. The truth is that the production of cellulosic ethanol has never passed the pilot unit phase, due to the lack of economic competitiveness. New ways must be found to make this viable. From the latest finding of the biomass structure, new biomass processing pathways are being advanced, constituting new biorefinery models, which will make it possible to obtain cellulosic ethanol concomitant with the production of different bioproducts such as xylitol, oligosaccharides, antioxidants and analogues to carbon fiber, etc. Lipid rich biomass is the source of foods oils. With population growth, the amounts of waste volume will increase. It is important to improve the processes of valorization of these residues, through their conversion into alcoholic esters of fatty acids, which can be used as fuel or in other domestic and industrial applications. This book focuses on the technologies developed for the conversion of all three biomass components, i.e. cellulose, hemicellulose and lignin, and their constituents, to fuels and high-value products. Both biochemical and thermochemical approaches are reviewed. Additionally, the developed technologies are described in detail and their potential applications as well as their commercial status are discussed. The early attempts to produce fuel ethanol from lignocellulosic biomass feedstock focused solely on the biological conversion of cellulose because the only organism that had been used successfully for commercial production of ethanol, i.e. Saccharomyces cerevisiae, could only ferment glucose, which was obtained from the hydrolysis of cellulose. Hemicellulose and lignin were considered as wastes in these processes and were normally removed in pretreatment processes to enhance enzymatic hydrolysis of the remaining cellulose. However, this approach was not economically feasible and as a result, the biorefinery concept was developed. In a biorefinery, in addition to ethanol, various higher-value products are produced from hemicellulose and lignin, which were previously not considered. Consequently, technologies were developed for the fractionation of biomass and conversion of hemicellulose and lignin to fuels and high-value products to improve the economic feasibility.
