Cell Surface GRP78, a New Paradigm in Signal Transduction Biology presents a new paradigm that has emerged in the past decade with the discovery that various intracellular proteins may acquire new functions as cell surface receptors. Two very prominent examples are ATP synthase and GRP78. While the role of cell surface ATP synthase has been reviewed in various books, this book directs its attention to the story of cell surface GRP78. Edited by the researcher who identified cell surface expression of the molecular chaperone GRP78 as a major factor in prostate cancer and other malignancies Presents an in-depth treatment of the biological underpinnings of GRP78 and its connection to disease Provides four-color illustrations that facilitate the narrative

Establishment of a normal phenotype involves dynamic epigenetic regulation of gene expression that when affected contributes to human diseases. On a molecular level, epigenetic regulation is marked by specific covalent modifications (acetylation, methylation, phosphorylation, sumoylation, PARylation and ubiquitylation) of DNA and its associated histones. Studies also suggest the influence of such epigenetic modifications on non-coding RNA expression implicated in normal and diseased phenotypes. Epigenetic control of genetic expression is a reversible process essential for normal development and function of an organism. Alteration of epigenetic regulation leads to various disease forms such as cancer, diabetes, inflammation and neuropsychiatric disorders. Assessing these alterations provides a deeper insight into the changes induced in the genome, which is often informative for identifying disease subtypes or developing suitable treatments. Therefore, epigenetics proves to be a key area of clinical investigation in diagnosis, prognosis, and treatment of complex diseases. Genetic mutations, environmental stress, pathogens and drugs of abuse are some of the predominant factors that induce and impact changes on chromatin, which directly dictate a diseased phenotype. It is essential to consider the interaction between genetic and epigenetic factors to understand the molecular mechanisms of complex human diseases for safer and efficient drug development. Furthermore, genetic variation in absorption, distribution, metabolism, and excretion (ADME) genes is insufficient to account for interindividual variability of drug response. Therefore, current efforts aim to identify epigenetic components of ADME gene regulation, which include phase-I and phase-II enzymes, uptake transporters, efflux transporters and nuclear receptors involved in regulation of ADME genes. Monitoring circulatory epigenetic biomarkers in liquid biopsies (blood, saliva, urine, cerebrospinal fluid) of disease-associated and drug-associated epigenetic alterations may prove useful for decision support for routine clinical treatment and drug discovery. Hence, recent drug discovery efforts on targeting the epigenome, has emerged an area of interest with several new drugs being developed, tested and some already approved by the US Food and Drug Administration (FDA). These new insights into the complexities of epigenetic regulation are key contributors to our basic understanding of this process in human health and disease, which will provide scope for innovative drug therapies. It is of urgency to aid the present understanding of epigenomics driven diseased outcomes, with the expectation that further studies will identify early markers of disease and targets for therapeutics.

Heat shock proteins are emerging as important molecules in the development of cancer and as key targets in cancer therapy. These proteins enhance the growth of cancer cells and protect tumors from treatments such as drugs or surgery. However, new drugs have recently been developed particularly those targeting heat shock protein 90. As heat shock protein 90 functions to stabilize many of the oncogenes and growth promoting proteins in cancer cells, such drugs have broad specificity in many types of cancer cell and offer the possibility of evading the development of resistance through point mutation or use of compensatory pathways. Heat shock proteins have a further property that makes them tempting targets in cancer immunotherapy. These proteins have the ability to induce an inflammatory response when released in tumors and to carry tumor antigens to antigen presenting cells. They have thus become important components of anticancer vaccines. Overall, heat shock proteins are important new targets in molecular cancer therapy and can be approached in a number of contrasting approaches to therapy.

This book is about Nutraceuticals in cancer therapy, specifically targeted and Adjuvant therapy. It shows several approaches for possibly reducing systemic toxicity. This book illustrates the role of several dietary agents, collectively called nutraceuticals or natural agents in the prevention and/or treatment of human malignancies known to be mediated through alterations in multiple molecular targets. This book contains sixteen chapters which begin with historical perspective on the value of natural agents in the prevention of human malignancies followed by a series of current topics on multiple nutraceuticals targeting multiple cancers. This collection would likely be useful for bringing newer generations with broader perspectives in launching cutting-edge innovative molecular research, which would certainly help in designing targeted clinical trials in order to realize the dream of customize strategies for the prevention and/or treatment of human malignancies without causing any systemic toxicity. Moreover, the knowledge gained would allow novel utilization of nutraceuticals as adjunct to both conventional chemotherapy and radiation therapy in order to improve the overall quality of life and survival of patients diagnosed with cancers.

An increasing amount of cancer research is being directed towards the investigation of plant-derived anticancer compounds, many of which have been used in traditional herbal treatments for centuries. Plants that Fight Cancer is an up-to-date, extensive review of plant genera and species with documented anti-tumor and anti-leukaemic properties. Following an overview of the disease and the diverse methods of therapy and clinical testing, the book provides a detailed examination of the plants whose compounds are currently used in conventional cancer treatment, the species which show the greatest potential as future candidates, and other species with established anticancer properties. The third section explores each of more than 150 terrestrial plant genera and species, with a review of their traditional uses, mythology, botany, active ingredients, and product applications, along with photographs and illustrations and an analysis of expected results and risks. The text closes with a discussion of algal extracts and isolated metabolites with anticancer activity, a summary of published research for each species, and chemical structures of the most important compounds.
