A tandem benzannulation-ring closing metathesis strategy for the efficient synthesis of benzo-fused nitrogen heterocycles such as dihydroquinolines, benzazepines, and benzazocines has been developed. This strategy is based on the benzannulation reaction of ynamides with cyclobutenones or [alpha]-diazo ketones to generate highly-substituted aniline derivatives, which then participate in ring-closing metathesis reactions to form nitrogen heterocycles. The synthetic utility of this strategy has been demonstrated by its successful application in a formal synthesis of the natural product (+)-FR900482. In addition, an environmentally-friendly approach to the synthesis of amides and lactams has been developed using supercritical carbon dioxide as a 'green' replacement solvent. The amide products are generated from the addition of amines to ketenes, which are formed in situ from the retro-ene reaction of alkynyl ethers.

Benzannulations employing ynamides and vinylketenes (generated in situ from [alpha]-diazo ketones) were investigated. Irradiation of the diazo ketones using a batch or continuous-flow reactor leads to the formation of vinylketenes via a photo-Wolff rearrangement. The vinylketenes then react with ynamides via a pericyclic cascade process to produce highly substituted aniline derivatives. Using this vinylketene-based benzannulation, tandem strategies for the synthesis of highly substituted benzo-fused nitrogen heterocycles were investigated. A tandem benzannulation-iodocyclization method for the synthesis of polysubstituted quinolines was established. In addition, a tandem strategy for the synthesis of carbazoles was developed and applied in the total synthesis of the carbazole alkaloid carazostatin as well as formal syntheses of the alkaloids carbazoquinocin C and antiostatin A4.

Highly convergent annulation strategies have been developed for the synthesis of azulenes and polycyclic nitrogen heterocycles. Specifically, substituted azulenes have been synthesized via a ring expansion-annulation strategy based on n-halo diazo ketones. The method employs the use of readily available benzenoid starting materials and achieves the synthesis of azulenes substituted on both the five- and seven-membered rings regioselectively. The synthetic utility of the ring expansion annulation products, in particular azulenyl halides and triflates, has been demonstrated by their successful application in transition metal-mediated carbon-carbon bond forming reactions. These reactions have been used to further functionalize the five- and seven-membered rings and also in the synthesis of oligoazulenes and azulenylamino acid derivatives. In addition, polycyclic nitrogen heterocycles have been synthesized via a tandem ring forming strategy. The strategy employs a novel benzannulation reaction of substituted cyclobutenones with ynamides, which are synthesized via the strategy for the N-alkynylation of amides strategy developed in the Danheiser laboratory. The products of these benzannulation reactions, highly-substituted anilines, then participate in ring closing metathesis reactions to form various nitrogen heterocycles. In particular dihydroquinolines, hydrobenzoazepines, and hydrobenzoazocines were synthesized via this tandem ynamide benzannulation ring closing metathesis strategy.
