Cellular targeting of drug delivery systems represents a promising approach to reduce adverse effects by limiting release of pharmacological agent to desired target site. Controlled surface modification of nanoparticles by specific ligands can facilitate maximum receptor binding and cellular internalization, thereby increasing efficacy of targeting. This research attempts to establish the foundation for ligand-mediated, covalent surface modification of prefabricated carboxyl functionalized polymeric nanoparticles. Control over size and surface charges affecting stability is demonstrated through fabrication of particles in the absence and presence of a stabilizer. Covalent conjugation chemistry for amidation of surface carboxyl groups to amine-bearing ligands is explored and challenges in separating unbound ligands without compromising particle stability are presented. Successful synthesis and purification of a transcytosis-facilitating peptide sequence ( -RYRGDLGRR- ) and its analogues are outlined. Lastly, increased transport of peptide-modified nanoparticles across BeWo cell monolayers, an in vitro model of the human placenta is demonstrated. Preliminary competition studies showing this increased transport of peptide-functionalized particles to be a result of receptor mediated interactions are put forth. In summary, we provide sufficient experimental evidence to suggest increased transport properties of modified particles to be dependent on covalent surface conjugation of appropriate ligands. This establishes the foundation to evaluate controlled expression of targeting ligands for optimizing targeting efficiency of delivery systems.

Nanotechnology has the potential to change every part of our lives. Today, nanotechnology-based products are used in many areas, and one of the most important areas is drug delivery. Nanoparticulate drug delivery systems not only provide controlled delivery of drugs and improved drug solubility but also improve drug efficiency and reduce side effects via targeting mechanisms. However, compared with conventional drug delivery systems, few nanoparticle-based products are on the market and almost all are nontargeted or only passively targeted systems. In addition, obtaining targeted nanoparticle systems is quite complex and requires several evaluation mechanisms. This book discusses the production, characterization, regulation, and currently marketed targeted nanoparticle systems in a broad framework. It provides an overview of targeted nanoparticles’ (i) in vitro characterization, such as particle size, stability, ligand density, and type; (ii) in vivo behavior for different targeting areas, such as tumor, brain, and vagina; and (iii) current advances in this field, including clinical trials and regulation processes.

This book is the first of its kind to offer a comprehensive and up-to-date discussion of the use of nanoscale materials for biomedical applications, with a particular focus on drug delivery, theragnosis and tissue regeneration. It also describes in detail the methods used in the preparation of nanoparticles. Response of nanoparticles in biological systems are also explored. Nanotechnology has led to the advent of a new field, nanomedicine, which focuses on the use of nanomaterials as drug-delivery vehicles to develop highly selective and effective drugs. The combination of molecular imaging and nanotechnology has produced theragnostic nanoparticles, which allow the simultaneous detection and monitoring of diseases. Nanotechnology can also be combined with biomaterials to create scaffolds for tissue regeneration. Further, significant advances have been made in the areas of drug delivery, theragnostic nanoparticles and tissue regeneration materials. Some nanomedicines and tissue regeneration materials are already commercially available, while others are undergoing clinical trials, and promising results have been documented. Despite the rapid advances in nanomedicine, there is a relative dearth of literature on the biomedical applications of nanoscale materials.
