This dissertation summarizes the development of novel synthetic methods involving organometallic reagents and their application in the total synthesis of the natural product eupomatilone 2. These newly discovered reactions including the palladium-catalyzed cross-coupling of allyl acetates with organosiloxanes and the diastereoselective synthesis of cis- and trans-a-methylene-y-lactones utilizing Baylis-Hillman adducts as precursors. Another important aspect of the research is the application of some of the methods in total synthesis of eupomtilone 2. The new synthetic methodologies involving organometallic reagesnts developed in this dissertation are important transformations in modern organic synthesis. Mild reaction conditions, tolerance of various organic functional groups, and high selectivity are advantages of these methodologies. The ability to recycle the catalytic system in some cases is another attractive feature for PEG-involved methods. The short and efficient strategy for the synthesis fo eupomatilone 2 could serve as a catalyst for research in this area.

These newly discovered reactions include the rhodium-catalyzed direct cross-coupling of allylic alcohols with aryl- and vinylboronic acids; palladium-catalyzed coupling reaction of Baylis-Hillman acetate adducts with both potassium organotrifluoroborate reagents and organosiloxanes; efficient syntheses of functionalized allylmetals such as allylboranes, allylsilanes, and allylgermanes; application of allylboranes to the synthesis of homoallylic alcohols; syntheses of 4-(1-alkynyl)-2(5H)-furanones and coumarins; utilization of potassium alkynyltrifluoroborates in the synthesis of conjugated enediynes; and application of potassium alkynyltrifluoroborates as reagents in the Petasis reaction. The new synthetic methodologies involving Group III & IV reagents developed in this dissertation are important transformations in modern organic synthesis. The development of novel synthetic methodology and the total synthesis of natural products are essential mainstays of organic chemistry. The development of novel synthetic methodology to accomplish new molecular transformations is of great importance to organic chemistry. As well, total synthesis of natural products commands significant interest because of the vast potential many natural products possess for beneficial medicinal application. In the exploration of natural products, total synthesis substantially empowers the researcher who seeks to reassign natural product structures that have been misassigned. This dissertation incorporates both novel synthetic methodology and total synthesis through the trajectory of three distinct projects. Chapter one describes the significant advances made in the carbon-carbon bond forming reaction produced by the direct displacement of alkoxy groups of vinylogous esters by Grignard reagents. Applications of this reaction have shed light on interesting carbonyl reactivity. Increased mechanistic understanding of this displacement reaction has been achieved through a meld of theoretical computation and physical experimentation. Chapter two addresses the significant progress made toward the total synthesis of the complex natural products chrysophaentins A and E. These molecules are compelling targets for investigation not only because of the novel antibiotic potential that chrysophaentin A features but also because of the natural products' structural uniqueness. Chapter three expounds on the progress made toward the total synthesis of erythroculine N-oxide with the goal of revising the likely misassigned reported structure of the natural product jamtine N-oxide.
